1. What is Karyometrix?

Stemnovate utilizes a digital karyotyping tool that screens and identifies structural chromosomal variations such as deletion, duplication, insertion and translocation changes on a genome-wide scale.

Typically high molecular weight DNA is isolated from the cell line and the genomic DNA (gDNA) is enzymatically digested and tagged with sequencing adaptors. Barcoded adapter-tagged g-DNAs are then transferred into a flow cell for accurate long-read sequencing and base-calling. Nucleotide raw reads are analyzed, assembled, and aligned against the reference genome to detect structural variations in both coding and non-coding regions of the chromosome. The Karyometrix platform will provide users with unprecedented access to crucial genetic information that will facilitate new therapeutic discoveries.

that is limited to SV’s > 500bps which in some cases could miss out key genetic events.

How does it work?

In this method, high molecular weight DNA is isolated from the cell line and the genomic DNA ( gDNA) is enzymatically digested and tagged with sequencing adaptors on the 5’ phosphate end and 3’ hydroxyl group end of the gDNA . Barcoded adapter tagged gDNAs are then transferred into a flowcell for accurate long read sequencing and basecalling. Raw reads which can be in either a FASTQ or BAM format are computationally analysed, assembled and aligned against the reference genome e.g. GRCh38 or any custom reference genome of choice to detect structural variations in both coding and non- coding regions of the chromosome.

G-banding

Cytogenetic G-banding is a technique used to produce a visible karyotype, grouping of all chromosomes, by staining condensed chromosomes. By photographing the entire complement of chromosomes, it helps identify genetic diseases. The metaphase chromosomes are treated with trypsin (which partially digest proteins that envelope chromosomes called histones) and Giemsa stain. More dark staining occurs in heterochromatic regions, which are highly adenine and thymine-rich and relatively gene-poor. The less condensed chromatin (Euchromatin), which is rich in guanine and cytosine and more transcriptionally active, incorporates less Giemsa stain. The pattern of bands is numbered from the centromere to the telomere on each arm of the chromosome. With this numbering system, any band on the chromosome can be identified and described precisely.
